I upgraded my iPhone 4S to it and have had no problems. But if you are a heavy Google Maps user, you may want to hold off until Apple's Maps matures a bit. But Apple's Maps does have turn by turn directions now and it does have a pleasing interface, overall. Lighter users of Maps will probably be just fine with it. One thing I did notice, though, is that it doesn't show estimated property lot lines on streets as the Google Maps app did.
It also has an autolock feature that will engage during calls. I turned this off, though I will likely use it when I travel (I use pass phrases to lock my phone).
The mail view is a bit different, but functionally the same overall. It does add the VIP designation for email from important sources (i.e., spouse, family, et al).
I like the privacy features for the phone and phone calls. You have some ability to screen and filter how phone calls from different people are handled by your phone, especially when you are busy on other calls. I am often on conference calls during the day, so this is a nice feature that you can schedule.
I haven't enabled the Facebook integration because I am not really interested in doing that. But some folks may find that as a convenience.
If you like the YouTube app, you will need to download that separately now, since it is gone in the native iOS installation.
Those are just my first impressions.

I upgraded to iOS6 as soon as it was available. No problems, no surprises, looks pretty much like the old OS. Lots of new features, especially for Facebook users. Better app store. No, you don't have Google maps at the moment, but you do have turn-by-turn nav now. Go for it.

I upgraded my iPad 2 & iPhone 4s, no problems with either except for minor glitches, such as needing to turn iMessage back on, and the Passbook app doesn't open the App Store.... There is a fix though, a wierd one, but found on the Internet and it works... Set the data to next year, then reopen Passbook... Then fix the date, and now Passbook will work. I know that makes no sense, but it fixed it. You can easily search that on the net.
One thing I found that. I love is the ability to delete a persons email from the auto type when sending an email. In other words, if you want to delete someone's old email address, it's easy to change the contact, but when you start typing an email, it would auto fill their old email... Now that can be deleted.
My only real issue is with the iCloud control panel app for Win 7. The photo stream doesn't work anymore. It used to do as advertised, upload pictures taken to the windows computer, now it does not, it always unchecks it self from the control panel app. You need to email pictures to a Win machine till they fix it.
I was only a 600mb download, so I think it's just new features added, some things tweaked and that's about it. Don't expect a major change like from iOS 4 to 5. 6 is just a few new things added.

I have to admit after using this a few days... I miss Streetview in Google Maps. I didn't realize how much I used that till today. It was really integrated nicely into that app.

I like my Tom Tom too...but I can see how I might be caught out without it in a rentacar or something. But that's just the tip of the iceberg. Push notificatons for severe weather from the Weather Channel app. Processing for the camera's images. Easy panoramas. Integraton of Facebook contacts. And on and on. SOMETIMES--not often but sometimes--there is such a thing as a free lunch.

We have a Tom Tom GPS, so it was a no brainer to get the Tom Tom iPad app. We're used to the UI. GPS's seem to have the worst UI's out there. 
Some of the Facebook stuff sounds like an improvement. Hmmmmm

Yep. Upgraded my 4th gen iPod and my iPhone 4. Will upgrade the GF's iPhone 4s today. With the exception of that early WiFi issue, no major surprises and both of my devices are running fine.
Apparently there are some pretty dense and spoiled Apple users out there. The Community forums are flooded with boo-hooing over not having Siri on the older devices.

Think Apples Maps will give Google a run for its' money; at some point (and all us users will benefit from this competition) but it has a ways to go. Google has a very good product.

Turn by turn directions on Maps seemed to work okay for me on the one trip where I tried it (I had my Garmin going at the same time and they both came up with the same route which took me around some highway construction).
A problem that I did run into is something that has been a bit of an issue with earlier iOS updates if you are using any push services (iCloud, Exchange, or any IMAP mail services) with the calendar. I found my calendar didn't have any calendars loaded and so it was blank of appointments. The solution was to check the push services and make sure they are turned on to push. If you are using manual push settings (which I do when I travel to save a bit on the batteries and control data usage overseas), you need to change the settings to push and then you can go back to manual.
I think that this may be happening because I do generally have my settings on manual for the push services. It is possible that when a new iOS is installed that it resets the push services control (which is separate, incidentally) and clears the calendar for some unknown random reason. This also happened when I upgraded to iOS 5.

Maybe Im being paranoid, but my wifes Iphone iOX5 using Google maps, gets our home location spot on, my ipad on iOX5 Google Maps - spot on, iOX6 Apple Maps - spot on, iOX6 Google Earth - about 30 metres off (all the time..)

Goto Apple maps in iOS 6, then the Hoover dam, turn on 3D, the road over the dam gets real bumpy, then move sightly to i93, thats going to one ride you won't forget, but you will still get similar situations in Google maps, its just Google has fixed up many problems
